L. b. (Lachnospiraceae bacterium) Cas12a Nuclease, also known as Cpf1, is an RNA-guided endonuclease derived from the Class II type V CRISPR-Cas system. It exhibits a high degree of specificity in targeting and cleaving particular DNA sequences in genome editing. Additionally, Cas12a nuclease possesses collateral cleavage activity, resulting in the non-specific cleavage of neighboring DNA upon target recognition.
The DNA cleavage capability of Cas12a nuclease has been leveraged for various applications, including genome editing, gene regulation, and nucleic acid detection. By programming Cas12a with specific guide RNAs, targeted modifications such as gene knockout, gene insertion, or point mutations can be introduced into the target DNA sequence.
