Glycogen is the principal storage form of glucose in animal and human cells. It is made primarily by the liver and muscles, but it can also be made by glycogenesis within the brain and stomach. Cayman’s Glycogen Assay provides a simple, reproducible, and sensitive tool for assaying glycogen from tissue. In this assay, glycogen is hydrolyzed by amyloglucosidase to form β-D-glucose, which is then specifically oxidized to D-glucono-δ-lactone by glucose oxidase forming hydrogen peroxide in the process. Hydrogen peroxide, in the presence of horseradish peroxidase, reacts with 10-acetyl-3,7-dihydroxyphenoxazine (ADHP) in a 1:1 stoichiometry to generate the highly fluorescent product resorufin which is measure at an excitation wavelength of 530-540 nm and an emission wavelength of 585-595 nm.

Cell Biolabs’ Glycogen Assay Kit (Fluorometric) is a simple assay that measures the amount of total glycogen present in biological samples in a 96-well fluorescence plate reader. Glycogen is broken down into glucose monomers by amyloglucosidase first, glucose is then oxidized by glucose oxidase into D-gluconic acid and hydrogen peroxide. The resulting hydrogen peroxide is then detected with a highly specific fluorometric probe in a standard 96-well colorimetric plate reader. Sample glycogen concentrations are determined by comparison with a known glycogen standard. The kit has a detection sensitivity limit of 120 nM glycogen. Each kit provides sufficient reagents to perform up to 100 assays, including blanks, glycogen standards, and unknown samples. Note each sample replicate requires 2 assays, one of which is a negative control to be subtracted as background.

Oxidative damage to DNA is a result of its interaction with reactive oxygen species (ROS), in particular the hydroxy radical. Hydroxy radicals, which are produced from superoxide anion and hydrogen peroxide by the Fenton reaction, produce multiple modifications in DNA. Oxidative attacks by hydroxy radicals on the deoxyribose moiety leads to the release of free bases from DNA, generating strand breaks with various sugar modifications and simple abasic sites (AP sites). The Oxygen Radical Antioxidant Capacity (ORAC) Assay is a classic tool for measuring the antioxidant capacity of biomolecules from a variety of samples. The ORAC Activity Assay is based on the oxidation of a fluorescent probe by peroxyl radicals by way of a hydrogen atom transfer (HAT) process. Peroxyl radicals are produced by a free radical initiator, which quenches the fluorescent probe over time. Antioxidants present in the assay work to block the peroxyl radical oxidation of the fluorescent probe until the antioxidant activity in the sample is depleted. The remaining peroxyl radicals destroy the fluorescence of the fluorescent probe.
